If you re-upload does it write over the old one?

I'm pretty sure it does, but don't take my word for it. ;)

I believe it does.

It overwrites it, but keep in mind that none of the changes from your previous upload save when you reopen the game. So if you want to change one thing, you have to re-do all your other changes in addition to that one thing.
